Cytidine 5'-monophosphate (CMP)-induced structural changes in a multifunctional sialyltransferase from Pasteurella multocida.
Sialyltransferases catalyze reactions that transfer a sialic acid from CMP-sialic acid to an acceptor (a structure terminated with galactose, N-acetylgalactosamine, or sialic acid). They are key enzymes that catalyze the synthesis of sialic acid-containing oligosaccharides, polysaccharides, and glycoconjugates that play pivotal roles in many critical physiological and pathological processes. Th...
متن کاملComplete switch from α-2,3- to α-2,6-regioselectivity in Pasteurella dagmatis β-D-galactoside sialyltransferase by active-site redesign.
Structure-guided active-site redesign of a family GT-80 β-D-galactoside sialyltransferase (from Pasteurella dagmatis) to change enzyme regioselectivity from α-2,3 in the wild type to α-2,6 in a P7H-M117A double mutant is reported. Biochemical data for sialylation of lactose together with protein crystal structures demonstrate highly precise enzyme engineering.
متن کاملA multifunctional Pasteurella multocida sialyltransferase: a powerful tool for the synthesis of sialoside libraries.
A multifunctional sialyltransferase has been cloned from Pasteurella multocida strain P-1059 and expressed in E. coli as a truncated C-terminal His6-tagged recombinant protein (tPm0188Ph). Biochemical studies indicate that the obtained protein is (1) an alpha2,3-sialyltransferase (main function), (2) an alpha2,6-sialyltransferase, (3) an alpha2,3-sialidase, and (4) an alpha2,3-trans-sialidase. ...
متن کاملFirst report of Pasteurella dagmatis isolation from a bitch urine in Iran
Pasteurella species are commensal bacteria of the respiratory airways and oronasal cavity of animals. In this study, we report an unusual case of Pasteurella dagmatis isolation from dog urine with uroliths. An 11-year-old female dog was admitted to the Veterinary Hospital of Ahvaz University complaining of abdominal pain, hematuria, dysuria and stranguria. In clinical and radiological examinati...
متن کاملProsthetic valve endocarditis caused by a Pasteurella dagmatis-like isolate originating from a patient's cat.
Pasteurella species are part of the oral flora of cats and dogs. In humans, they are frequently found in infected animal bite wounds, but invasive infections are rare. This is the first report of prosthetic-valve endocarditis with a Pasteurella dagmatis-like species, which originated from the patient's cat.
